"Single Cell Anniversary" is the second segment of the twenty-second episode of season 6, and it is paired with "Chum Bucket Supreme." In this episode, SpongeBob helps Plankton find an anniversary gift for Karen.

Plankton enters the Chum Bucket, stinking with the stench of failure, so Karen cleans him. Afterwards, she tells him it's their anniversary and that her gift to him is a copy of the Krabby Patty secret formula. However, she tells him he can't have it until he gets a gift from his heart.

SpongeBob overhears Plankton while walking with Squidward. He tells him about his wedding anniversary and how he hadn't gotten anything for Karen yet. SpongeBob tells him he should get a gift from his heart as that is where love grows, and teaches him how to spell love.

At SpongeBob's house, he asks Plankton about what he and Karen like to do and the characteristics and hobbies of Karen. When he learns Karen makes a ding sound once a day, SpongeBob tells him he should sing for her. He teaches him how to sing, but his singing is extremely loud. Despite that, SpongeBob thinks he's ready.

They go to the Chum Bucket, where Plankton sings his song to Karen with SpongeBob and Patrick playing instruments as his anniversary gift to her. After finishing, SpongeBob realizes Plankton will get the formula in return for singing the song, and tries to turn off Karen before she loads the formula, but fails.

However, he still doesn't get the formula as Karen explodes due to the happiness she felt from the song. E.M.I.L.P., Karen's mother, loads and scolds Plankton for what he caused to her daughter.
